2-三氟甲基苯并咪唑盐作为苯甲醇脱氧三氟甲基化的双功能试剂

近日,上海大学张志坤团队报道了2-三氟甲基苯并咪唑盐作为苯甲醇脱氧三氟甲基化的双功能试剂。2025年10月22日,《中国化学》杂志发表了这一成果。

由于醇的普遍性和易接近性,醇的脱氧三氟甲基化是构建C(sp3)-CF3键的一种极具吸引力的策略。虽然传统方法依赖于活化/三氟甲基化过程,但需要单独的试剂进行醇活化和CF转移。相反,醇的脱氧氟化试剂通常将活化和氟化结合在一起。类似的用于脱氧三氟甲基化的双功能试剂尚未开发。

在这项研究中,研究组证明了2-三氟甲基苯并咪唑盐作为一种双重功能试剂,能够在脱氧三氟甲基化反应中同时激活苯甲醇和递送三氟甲基。

Title: 2-Trifluoromethyl-Benzimidazolium Salt as a Dual-Function Reagent for Deoxytrifluoromethylation of Benzyl Alcohols

Abstract: Deoxytrifluoromethylation of alcohols represents a highly attractive strategy for constructing C(sp3)-CF3 bonds owing to the ubiquity and ready accessibility of alcohols. While conventional approaches rely on activation/trifluoromethylation process, requiring separate reagents for alcohol activation and CF transfer. In contrast, reagents for deoxyfluorination of alcohols often integrate activation and fluorination together. Similar dual-function reagents for deoxytrifluoromethylation remain underdeveloped. In this study, we demonstrate the use of 2-trifluoromethyl-benzimidazolium salt as a dual-function reagent capable of simultaneously activating benzyl alcohols and delivering the trifluoromethyl group in deoxytrifluoromethylation reactions.
