科学家利用大鼠多能干细胞获得功能性原始生殖细胞样细胞

日本国立生理科学研究所Toshihiro Kobayashi、Masumi Hirabayashi等研究人员合作利用大鼠多能干细胞获得功能性原始生殖细胞样细胞。该项研究成果发表在2022年4月7日出版的《科学》杂志上。

研究人员表示,从多能干细胞(PSC)体外生成生殖细胞,对未来的生殖医学和动物育种有实质性的影响。十年前,体外配子发生在小鼠身上得到了确立。然而,诱导原始生殖细胞样细胞(PGCLC)产生配子还没有在任何其他物种中实现。

研究人员展示了从大鼠PSC中诱导出功能性PGCLC。结果表明,浮动聚集的外胚层细胞样细胞形成大鼠PGCLC。性腺体细胞支持PGCLC的成熟和表观遗传学的重编程。当大鼠PGCLC被移植到无精子大鼠的生精小管中时,会产生功能性精子,即那些能够孕育有活力后代的精子。从这个大鼠模型中得到的启示,将阐明体外配子发生的广泛适用性所必需的保守和不同的机制。

Abstract: The in vitro generation of germ cells from pluripotent stem cells (PSCs) can have a substantial effect on future reproductive medicine and animal breeding. A decade ago, in vitro gametogenesis was established in the mouse. However, induction of primordial germ cell–like cells (PGCLCs) to produce gametes has not been achieved in any other species. Here, we demonstrate the induction of functional PGCLCs from rat PSCs. We show that epiblast-like cells in floating aggregates form rat PGCLCs. The gonadal somatic cells support maturation and epigenetic reprogramming of the PGCLCs. When rat PGCLCs are transplanted into the seminiferous tubules of germline-less rats, functional spermatids—that is, those capable of siring viable offspring—are generated. Insights from our rat model will elucidate conserved and divergent mechanisms essential for the broad applicability of in vitro gametogenesis.
