CaMKII介导RGC可保护视力

美国西奈山伊坎医学院陈波研究组提出CaMKII介导的视网膜神经节细胞 (RGC)可保护视力。2021年7月22日出版的《细胞》发表了这项成果。

他们报告 CaMKII-CREB信号在 RGC 胞体的兴奋性毒性损伤或 RGC 轴突的视神经损伤后受到损害,并且该通路的重新激活有力地保护了 RGC 免受这两种损伤。CaMKII 活性还促进了正常视网膜中 RGC 的存活。此外,在两种青光眼模型中,CaMKII 的重新激活可保护 RGC,其中 RGC 因眼内压升高或遗传缺陷而退化。最后,CaMKII 重新激活在体内保护长距离 RGC 轴突投影并保留视觉功能,从视网膜到视觉皮层,以及视觉引导行为。

Title: Preservation of vision after CaMKII-mediated protection of retinal ganglion cells

Abstract: Retinal ganglion cells (RGCs) are the sole output neurons that transmit visual informationfrom the retina to the brain. Diverse insults and pathological states cause degenerationof RGC somas and axons leading to irreversible vision loss. A fundamental questionis whether manipulation of a key regulator of RGC survival can protect RGCs from diverseinsults and pathological states, and ultimately preserve vision. Here, we report thatCaMKII-CREB signaling is compromised after excitotoxic injury to RGC somas or opticnerve injury to RGC axons, and reactivation of this pathway robustly protects RGCsfrom both injuries. CaMKII activity also promotes RGC survival in the normal retina.Further, reactivation of CaMKII protects RGCs in two glaucoma models where RGCs degeneratefrom elevated intraocular pressure or genetic deficiency. Last, CaMKII reactivationprotects long-distance RGC axon projections in vivo and preserves visual function, from the retina to the visual cortex, and visuallyguided behavior.
