研究绘制出COVID-19尸检样本中不同器官的蛋白质组学图谱

2021年1月9日,西湖大学郭天南等研究人员合作在《细胞》杂志发表论文,该研究绘制出COVID-19尸检样本中不同器官的蛋白质组学图谱。

研究人员报道了19个COVID-19患者中来自七个器官144个尸检样品的蛋白质组学分析。研究人员对这些样品中的11394种蛋白质进行了定量,其中COVID-19患者与对照组相比有5336种蛋白质被干扰。这些数据显示,COVID-19患者的肺组织蛋白酶L1而非ACE2显著上调。在多个器官中检测到全身性过度炎症以及葡萄糖和脂肪酸代谢的失调。

研究人员还观察到COVID-19患者多个器官中涉及缺氧、血管生成、血液凝固和纤维化的关键因素失调。睾丸损伤的证据包括Leydig细胞的减少、胆固醇生物合成和精子活动的抑制。

Abstract: The molecular pathology of multi-organ injuries in COVID-19 patients remains unclear, preventing effective therapeutics development. Here, we report a proteomic analysis of 144 autopsy samples from seven organs in 19 COVID-19 patients. We quantified 11,394 proteins in these samples, in which 5336 were perturbed in the COVID-19 patients compared to controls. Our data showed that cathepsin L1, rather than ACE2, was significantly upregulated in the lung from the COVID-19 patients. Systemic hyperinflammation and dysregulation of glucose and fatty acid metabolism were detected in multiple organs. We also observed dysregulation of key factors involved in hypoxia, angiogenesis, blood coagulation and fibrosis in multiple organs from the COVID-19 patients. Evidence for testicular injuries include reduced Leydig cells, suppressed cholesterol biosynthesis and sperm mobility. In summary, this study depicts a multi-organ proteomic landscape of COVID-19 autopsies that furthers our understanding of the biological basis of COVID-19 pathology.
