比利时鲁汶大学Adrian Liston研究团队发现,CD4 T细胞辅助小胶质细胞成熟。2020年7月22日,《细胞》在线发表了这一成果。

研究人员联合使用成像、单细胞和手术方法来鉴定了小鼠和人脑中的CD69+CD4 T细胞群,这不同于循环的CD4 T细胞。驻留在大脑中的群体是通过活化的循环细胞从原位分化而来的,并由自身抗原和外周微生物组形成。
 
单细胞测序显示,在没有小鼠CD4 T细胞的情况下,常驻小胶质细胞仍然游离在胎儿和成年状态之间。这种成熟缺陷导致过度的未成熟神经元突触和行为异常。这些结果阐明了CD4 T细胞在大脑发育中的作用以及免疫和神经系统进化之间的潜在联系。

Abstract: The brain is a site of relative immune privilege. Although CD4 T cells have been reported in the central nervous system, their presence in the healthy brain remains controversial, and their function remains largely unknown. We used a combination of imaging, single cell, and surgical approaches to identify a CD69+ CD4 T cell population in both the mouse and human brain, distinct from circulating CD4 T cells. The brain-resident population was derived through in situ differentiation from activated circulatory cells and was shaped by self-antigen and the peripheral microbiome. Single-cell sequencing revealed that in the absence of murine CD4 T cells, resident microglia remained suspended between the fetal and adult states. This maturation defect resulted in excess immature neuronal synapses and behavioral abnormalities. These results illuminate a role for CD4 T cells in brain development and a potential interconnected dynamic between the evolution of the immunological and neurological systems.
