科学家实现对新冠病毒突刺蛋白人源单克隆抗体的快速分离与鉴定

美国范德比尔特大学医学中心James E. Crowe、Robert H. Carnahan等研究人员合作,实现了对新冠病毒(SARS-CoV-2)突刺蛋白人源单克隆抗体的快速分离与鉴定。这一研究成果于2020年7月10日在线发表在《自然—医学》上。

研究人员使用了快速抗体发现平台来分离数百种靶向SARS-CoV-2突刺(S)蛋白的人类单克隆抗体(mAb)。基于它们与S蛋白亚结构域的反应性以及与SARS-CoV的交叉反应性,研究人员将这些单克隆抗体分为五个主要类别。

这些单克隆抗体中有许多会抑制真正的SARS-CoV-2病毒的感染,大多数中和的单克隆抗体会识别S的受体结合域(RBD)。这项工作确定了SARS-CoV-2 S上的易感位点,并证明了这一抗体发现平台的速度和稳定性 。

Abstract: Antibodies are a principal determinant of immunity for most RNA viruses and have promise to reduce infection or disease during major epidemics. The novel coronavirus SARS-CoV-2 has caused a global pandemic with millions of infections and hundreds of thousands of deaths to date1,2. In response, we used a rapid antibody discovery platform to isolate hundreds of human monoclonal antibodies (mAbs) against the SARS-CoV-2 spike (S) protein. We stratify these mAbs into five major classes on the basis of their reactivity to subdomains of S protein as well as their cross-reactivity to SARS-CoV. Many of these mAbs inhibit infection of authentic SARS-CoV-2 virus, with most neutralizing mAbs recognizing the receptor-binding domain (RBD) of S. This work defines sites of vulnerability on SARS-CoV-2 S and demonstrates the speed and robustness of advanced antibody discovery platforms.
