美国再生元制药公司Christos A. Kyratsous等研究人员发现,SARS-CoV-2突刺蛋白的抗体混鸡尾酒疗法可防止单个抗体引起的快速突变逃逸。这一研究成果于2020年6月15日在线发表在《科学》上。

研究人员测试了四种有效中和SARS-CoV-2刺突蛋白的抗体抗性,无论是单独还是与鸡尾酒疗法混合使用时。这些抗体对人类中出现的突刺变体仍然有效。但是,在单个抗体的存在下进行体外传代后,新的突刺突变体迅速出现,导致中和作用的丧失。
 
这种逃逸也发生在结合刺突蛋白不同但重叠区域的抗体组合中。重要的是,用非竞争性抗体鸡尾酒疗法处理后不会产生逃逸突变体。

Title: Antibody cocktail to SARS-CoV-2 spike protein prevents rapid mutational escape seen with individual antibodies

Abstract: Abstract Antibodies targeting the spike protein of SARS-CoV-2 present a promising approach to combat the COVID19 pandemic; however, concerns remain that mutations can yield antibody resistance. We investigate the development of resistance against four antibodies to the spike protein that potently neutralize SARS-CoV-2, individually as well as when combined into cocktails. These antibodies remain effective against spike variants that have arisen in the human population. However, novel spike mutants rapidly appeared following in vitro passaging in the presence of individual antibodies, resulting in loss of neutralization; such escape also occurred with combinations of antibodies binding diverse but overlapping regions of the spike protein. Importantly, escape mutants were not generated following treatment with a non-competing antibody cocktail.
