科研人员开发近红外II窗口光学片显微镜

该课题组研究人员开发了一种新的近红外II区(1,000 -1,700 nm)光片显微技术,其激发和发射波长分别高达约1,320 nm和1,700 nm。在未进行侵入性手术的活体组织内,其成像穿透深度约750微米,在甘油清除的脑组织内,成像穿透深度约2mm。该近红外II区光片显微技术在法向和斜向配置下,通过完整的组织实现活体小鼠体内成像,揭示了肿瘤微循环中的异常血液流动和T细胞运动,在细胞分辨率水平上对肿瘤中的程序性死亡配体1(PDL-1)和程序性细胞死亡受体1(PD-1)分布进行了绘制。另外,研究者通过对完整的小鼠头部进行三维成像,可以有效辨别颅骨和大脑皮层之间的血管通道,从而监测创伤性脑损伤部位巨噬细胞和小胶质细胞的募集情况。

研究人员表示,由于光散射的存在,对哺乳活体动物进行非侵入性的高时空分辨深度组织三维光学成像具有很大的挑战性。

Title: Light-sheet microscopy in the near-infrared II window

Abstract: Non-invasive deep-tissue three-dimensional optical imaging of live mammals with high spatiotemporal resolution is challenging owing to light scattering. We developed near-infrared II (1,0001,700nm) light-sheet microscopy with excitation and emission of up to approximately 1,320nm and 1,700nm, respectively, for optical sectioning at a penetration depth of approximately 750m through live tissues without invasive surgery and at a depth of approximately 2mm in glycerol-cleared brain tissues. Near-infrared II light-sheet microscopy in normal and oblique configurations enabled in vivo imaging of live mice through intact tissue, revealing abnormal blood flow and T-cell motion in tumor microcirculation and mapping out programmed-death ligand 1 and programmed cell death protein 1 in tumors with cellular resolution. Three-dimensional imaging through the intact mouse head resolved vascular channels between the skull and brain cortex, and allowed monitoring of recruitment of macrophages and microglia to the traumatic brain injury site.
