研究人员报道了一种用于超分辨率荧光显微镜的干涉式单分子定位方法。荧光分子通过干涉条纹多个激发图案的强度进行定位,与具有相同光子强度的常规成像相比,该方法的定位精度提高了大约两倍。通过在25×25μm2的大范围视场中分辨尺寸低至5nm的纳米结构,研究人员演示了该技术。

Abstract: We introduce an interferometric single-molecule localization method for super-resolution fluorescence microscopy. Fluorescence molecules are located by the intensities of multiple excitation patterns of an interference fringe, providing around a twofold improvement in the localization precision compared with the conventional imaging with the same photon budget. We demonstrate this technique by resolving nanostructures down to 5nm in size over a large 25×25μm2 field of view.
